Dr. Peraramelli's research was focused on understanding the role of anti-coagulation proteins involved in regulating blood coagulation. In this book, role of TFPI in inhibiting extrinsic coagulation pathway has been thoroughly studied using enzyme kinetics. One of the major conclusions is that all the kunitz domains, N- and C-terminus are essential and important towards the inhibition. While the importance of C-terminus in binding to phospholipids and hence significantly contributing towards the inhibitory activity of TFPI is already known, the prime addition made by Dr. Peraramelli is to elucidate the role of the three kunitz domains of TFPI. The main topics discussed in the book are: 1) the role of three kunitz domains in the inhibition of FXa and TF-FVIIa; 2) binding of the cofactors, modulators to TFPI and their effect on the inhibitory activity of TFPI. Bleeding disorders such as Hemophilia could be treated by inhibiting TFPI. Information from this book could be a great source for producing effective TFPI inhibitors and hence one step closer to find potential therapies for patients with blood coagulation disorders.
